Impact/Purpose:

The purpose of this presentation is to highlight the available targeted bioactivity assay data relevant to the published thyroid adverse outcome pathway network, highlight the assay principles for some of these assays and indicate potential sources of assay interference and the general lack of orthogonal and confirmatory data. In doing so, this presentation indicates the need for use of these data in specific applications, such as contributing to weight-of-evidence, mechanistic hypotheses, and prioritization. Understanding these bioactivity data within an appropriate internal dose context, i.e., the maternal-fetal perinatal dose context, will enable the use of a bioactivity:exposure ratio relevant for selection of chemicals for screening in models of greater biological complexity.

Description:

Thyroid-relevant bioassay data in the publicly available ToxCast database (and browseable in the CompTox Chemicals Dashboard) will be reviewed by molecular-initiating event. Example case studies of how to interpret the suite of bioactivity assay data will be shown. Work-in-progress on understanding thyroid-relevant bioactivity in the maternal-fetal dose context will be highlighted, given that this perinatal context is likely most important for potential thyroid-related perturbation of neurodevelopment.
